A model of growth of icosahedral viral capsids is proposed. It takes into account the diversity of hexamers' compositions, leading to definite capsid size. We show that the observed yield of capsid production implies a very high level of self-organization

Capsidsareavitalpartofvirallifecycle,protectingitsmostessentialandthemostvulnerablepart,whichistheirgeneticcode.WeneedtoconsiderthatvirusescanhaveDNAorRNAgenomesandthelatterareespeciallysensitivetodegradation(bynucleases).Inotherwords,withoutthecapsid,thegenomesexposedtothehostileexternalworldwouldbedestroyedinacoupleofhoursbytheultravioletradiation,orbychemicalattacksofSO2,ozone03,NO2,etc.Thee cientprotectionprovidedbycapsidshellensureslongevityandmakespossiblelongtravelsfromonehosttoanother.Shouldweknowhowtohindercapsidassembly,tisinformationwouldhaveenormouspotentialbene tforthedevelopmentofantiviraltherapies.

Theexistenceofawell-de nedagglomerationschemeduringthecapsidproductionsuggestsmanypossibilitiesofdestructiveintervention.Asamat-teroffact,themorecomplicatedthesystemandthemoreintricatethelawsofitsfunctioning,themoretherearewaysofhinderingit.Letusconsiderafewexamplesofhowtodecreasethee ciencyofcapsidbuilding.

1)Createseven-sidedpolygons-heptamers-withthesameproteinsthatusuallyformpentamers.Suchaunitwillbealsosurroundedbyhex-amers,creatinglocalnegativecurvature,thusenhancingthepropercapsidproduction.Suchanachievementisquiteunlikelybecausofthesterichin-drance-thereisjustnotenoughplaceinordertopacksevenproteinswhichusuallygoinpacksby ve.Eveniftherewassuchapossibility,thehep-tamersshouldbecreatedinsidetheinfectedcells,ordeliveredthereinsomeway,whichisextremelydi cult.

2)Abetterwaytohindercapsidproductionwouldbetoproducehex-amersorpentamerswith“wrong”proteinsinserted,orinawrongorder-e.g.(abbaab)or(aaabbb)insteadof(ababab)sothattheycould twithoneside,butthenpresentawrongproteintonextcapsomerstryingtoagglom-erate,thusdestroyingthesymmetryandorderoftheconstruction.Again,thissupposesthecreationofmodi edRNAchainsorderingtheproductionofdi erenthexamers,andagain,thedeliveryproblemseemsverydi culttosolve.
